Pentadecapeptide BPC 157 (PL 14736) Improves Ligament Healing in the Rat — VialBase Research
BPC-157 significantly accelerated medial collateral ligament healing in rats
- BPC-157 significantly accelerated medial collateral ligament healing in rats
- Improved biomechanical properties (load to failure, stiffness)
- Enhanced collagen organization and type I collagen expression
- Effective via both local injection and systemic administration
- No adverse effects observed
Summary
This controlled animal study examined BPC-157’s effects on medial collateral ligament (MCL) healing in a rat transection model. Rats received either BPC-157 (10 mcg/kg intraperitoneally or locally) or saline control after MCL transection. Healing was assessed at multiple time points for biomechanical properties, histological organization, and gene expression.
Key Findings
- BPC-157-treated ligaments showed significantly greater load to failure and stiffness at 14 and 28 days post-injury
- Histological assessment showed improved collagen fiber organization and alignment
- Increased type I collagen expression (the mature, load-bearing collagen type)
- Improved vascularity at the healing site (consistent with BPC-157’s angiogenic properties)
- Both systemic (IP injection) and local administration were effective
- No adverse effects, toxicity, or abnormalities observed in treated animals
- Dose used (10 mcg/kg) is within the commonly extrapolated human dosing range
Relevance to BPC-157
This is one of the most frequently cited BPC-157 studies and provides some of the strongest biomechanical evidence for its healing properties. The demonstration of improved load-to-failure (the ligament’s ability to withstand force) is clinically meaningful — it’s not just faster healing but functionally superior healing. This study is often referenced in the context of stacking BPC-157 with TB-500 for sports injuries and post-surgical recovery. The finding that both local and systemic routes work supports the subcutaneous injection protocols commonly used.
